1) Complementation analysis of the Rhg4 gene: hairy root and stable soybean transformation under the control of native promoter. This experiment will unequivocally demonstrate that we have isolated the gene conferring resistance to SCN at the Rhg4 locus.
2) Characterize the Rhg4 gene: gene function, regulation and expression upon SCN infection. These experiments will help us determine exactly how this gene is functioning to confer resistance against SCN. 3) Investigate interactions between the Rhg4 gene and soybean genes/products. This experiment will enable us to identify additional soybean genes that may be functioning in the Rhg4-mediated resistance signaling pathway.
4) Test the function of potential soybean genes found to interact with the Rhg4 gene. These experiments will confirm additional plant components of the Rhg4-mediated signaling pathway.
5) Investigate interactions between the Rhg4 gene and nematode genes/products. This experiment will help us identify which genes in the nematode encode proteins that are detected by resistant soybean plants to trigger the resistance response.
6) Test the function of potential nematode genes found to interact with the Rhg4 gene. These experiments will aid in identifying the proteins used by virulent SCN to overcome or evade the Rhg4-mediated resistance response.
