In the first three years, we will collaborate with breeders to select 21 major germplasm used in all soybean oil breeding programs in the US, and construct a mixture of DNA libraries for each germplasm by applying a variety of library construction and DNA-barcode strategies, then sequence the libraries using Solexa sequence-by-synthesis technology. Meanwhile, we will determine lipid profile including oleic acid, linolenic acid and stearic acid levels, and total oil content in seeds of the 21 germplasm. We will further use bioinformatic tools to identify gene sequence and expression variations among 21 germplasm, and conduct association studies to identify putative functional markers for oleic acid, linolenic acid, stearic acid and total oil contents in those germplasm.
